چکیده

Nasopharyngeal carcinoma (NPC) is a malignant tumor originating in the epithelium. Radiotherapy is the standard therapy, but tumor resistance to this treatment reduces the 5-year patient survival rate dramatically. Studies are urgently needed to elucidate the mechanism of NPC radioresistance. Epigenetics—particularly microRNAs (miRNA) andDNAmethylation—plays an important role in carcinogenesis and oncotherapy. We used qRT-PCR analysis and identified an miRNA signature from differentially expressed miRNAs. Our objectives were to identify the role of miR24 in NPC tumorigenesis and radioresistance and to identify the mechanisms by which miR24 is regulated. We found that miR24 inhibited NPC cell growth, promoted cell apoptosis, and suppressed the growth of NPC xenografts. We showed that miR24 was significantly downregulated in recurrent NPC tissues. When combinedwith irradiation, miR24 acted as a radiosensitizer in NPC cells. One of themiR24 precursorswas embedded in aCpG island.AberrantDNAmethylationwas involved in NPC response to radiotherapy, which linked inactivation of miR24 through hypermethylation of its precursor promoter with NPC radioresistance. Treating NPC cells with the DNA-hypomethylating agent 5-aza-20deoxycytidine compensated for the reducedmiR24 expression. Together, our findings showed thatmiR24was negatively regulated by hypermethylation of its precursor promoter in NPC radioresistance. Our findings defined a central role for miR24 as a tumor-suppressive miRNA in NPC and suggested its use in novel strategies for treatment of this cancer. Mol Cancer Ther; 1–12. 2014 AACR.

Cellular response to ionizing radiation: A microRNA story

MicroRNAs (miRNAs) represent a class of small non-coding RNA molecules that regulate gene expression at the post-transcriptional level. They play a crucial role in diverse cellular pathways. Ionizing radiation (IR) is one of the most important treatment protocols for patients that suffer from cancer and affects directly or indirectly cellular integration. Recently it has been discovered that mi...

Study of promoter CpG island hypermethylation of cyclindependent kinase inhibitor gene p21waf1/cip1 on some breast carcinoma cell lines

The p21 belongs to the CIP/KIP family of CDK inhibitors involved in cell cycle arrest at specific stages of the cell cycle progression. DNA methylation is the best studied epigenetic mark that have been evidently associated to chromatin condensation, and repression of gene transcription. The CpG island hypermethylation in promoter region of certain genes occurs in cancer cells and affects tumor...

Comparative proteomics analysis of a novel g-radiation-resistant bacterium wild-type Bacillus megaterium strain WHO DQ973298 recovering from 5 KGy g-irradiation

In order to examine radiation-induced proteins in an extremely radio-resistant bacterium, it became possibleto perform comparative proteomic analysis on radio-resistance Bacillus megaterium WHO as a wildtypestrain for the first time. Variation in cellular proteins profiles of the Bacillus megaterium WHO after 5KGy γ-irradiation were analyzed by two-dimensional poly acryl amide...

Augmented expression levels of lncRNAs ecCEBPA and UCA1 in gastric cancer tissues and their clinical significance

Objective(s): As the second cause of cancer death, gastric cancer (GC) is one of the eminent dilemmas all over the world, therefore investigating the molecular mechanisms involved in this cancer is pivotal. Unrestricted proliferation is one of the characteristics of cancerous cells, which is due to deficiency in cell regulatory systems. Long non-coding RNAs (lncRNAs) have emerged as critical re...
